Detection of transgene expression in human bone marrow aspirates via rAAV-mediated gene transfer. Aspirates were co-transduced with rAAV-hTGF-β/rAAV-FLAG-hsox9 (10 or 20 μl each vector), transduced with rAAV-lacZ (20 or 40 μl), or let untreated for over time (21 days) maintenance in chondrogenic medium as described in the Materials and Methods. The samples were processed after 21 days to detect (a) the expression of β-gal, TGF-β, SOX9, and the FLAG tag by immunohistochemistry (original magnification x20, bar = 100 μm, insets at x40, all representative data) and (b) the production of TGF-β by ELISA as described in the Materials and Methods. *,**Statistically significant compared with control groups (rAAV-lacZ, no vector condition) (*p ≤ 0.050, **p ≤ 0.010)
